size exclusion or gel filtration main disadvantages are @1. there should be at least 10% difference in molecular masses to have a good resolution otherwise bands will not separate properly. on the other hand, precipitation is used in order to concentrate and purify proteins from various contaminants. size exclusion is time taken.

Size exclusion chromatography is a method of separating proteins and other molecules based on their size. Small molecules flow through the pore of the matrix and fit better than large molecules. Larger molecules take longer to move up the chromatography paper or TLC plate, whereas smaller molecules are more mobile. Likewise, the polarity of the molecules can affect how far the spots travel, depending on the type of solvent used.
