Fragmentation of the protonated or de-protonated molecular ions generated from ESI is generally limited, and the mass spectra are relatively simple. However, multiple-protonation of proteins and peptides occurs in the ESI process, and the ESI mass spectra might become more complicated. A single protein will show its characteristic cluster ions containing multiple-charged ions. The number of charges on the protein molecules will depend on the molecular weight of the protein and the number of accessible basic sites.
