why do the carbohydrate rich protein and membrane proteins does not obey the empirical relationship in SDS PAGE?

    SDS mainly linearized the protein and give a overall negative charge, so SDS PAGE separate on the basis of mol. mass only. Glycoproteins and membrane proteins will be denatured, bcz SDS is a very effective solubilizing agent. One other difference is normally globular protein have hydrophobic interactions in the interior of protein but membrane proteins need to interact with lipids so they are having hydrophobic residues over their surfaces. they are mostly receptors and channel proteins and glycoproteins are having important functions like antibodies so If you treat them with SDS their functions may lost due to denaturation, and then they will be of no use.
