why does RNA primer use instead of DNA primer in the process of dna replication

    Because in DNA replication the polymerization not initiate the denovo strand of nucleotide to overcome this lack the RNA primer is used. DNA pol needed 3'OH for addtion the new nucleotide

    Everyone has already answered that DNA primers are not available inside the cells which is the reason why at all priming is required for DNA synthesis. Without a primer template junction along wit with a free 3'-OH, DNAP can't synthesise de novo. DNAP catalyses a SN2 nucleophilic attack of by 3' OH on the alpha phosphate of the incoming complementary nucleotide. Thus it needs a primer to begin with. Whereas, RNAP can polymerise de novo. So initial priming is done by RNAP, which is later removed by DNAP I by its exonuclease activity which allows it to do nick translation. As the DNA ligase doesn't seal the nick between the RNA primer and the DNA, that nick can be used by DNAP to replace the RNA primer. But for PCR or any other in vitro assay, where we order primers, we use DNA. As Taq Pol or any other PCR polymerase can efficiently polymerise irrespective of DNA or RNA primer. Moreover, handling RNA is much more difficult owing to the versatile presence of RNases which limits its stability.

    hybrids are more stable so a DNA -RNA hybrid start new replica. and second reason is DNA pol. unlike RNA polymerase can not start new strand biosynthesis it can only add dNTPs ( energy poor in comparison of NTPs) to existing 3'.

    cause rna can be made autonomously i.e. it does not need any primer either to ...but in case of dna synthesis i.e. replication primer is necessary. So to make dna primer system have to replicate dna with pre existing dna primer. But in case of RNA transcription no pre existing primer is not necessary.
