10. Circular DNA of a bacterium was allowed to replicate once. In this experiment, some of the nucleotides were fluorescently labeled. The primase was able to accept the labeled as well as the unlabeled nucleotides. However, the DNA polymerase could use only the unlabeled nucleotides. The fluorescent label is found in (1) both the template and newly made strands (2) only the newly made strand (3) only the template strand (4) neither of the two strands (IT JAM BT 2012) Tashu

In the given case, the Escherichia coli is a mutant for DNA ligase hence, the okazaki fragments formed on a lagging strand will not join. ● In the second generation they form low molecular weight fragments and cannot undergo further replication. ● The leading strand continues to replicate in every generation. ● Therefore two peaks will be formed. ● The newly synthesised continuous DNA fragment formed on the leading strand will show a peak of high molecular weight. ● The okazaki fragments formed on the lagging strand will show a peak of low molecular weight.
