    After being synthesized with the Rough Endoplasmic Reticulum, the protein gets translocated to the interior lumen of the RER with the help of a signaling peptide. This signaling peptide gets cleaved later on thus producing a truncated functional protein. The signaling peptide usually lies on the N-terminal of the protein. So the correct option is '4 kDa peptide from the N-terminus'.
