(a) (D)18 15, The 50 uL of component E.coli cells (109 cfu/mL) were transformed using 0.5 mg of a 5 kb plasmid DNA to which 950 uL of soc medium was added. Only 50 uL of this was plated on a selective agar plate. After an 12 h incubation at 37°C, 90 colonies were observed. Calculate the percentage in transformed cells. (a) 0.36% (b) 0.72% (C) 3.6%% (d) 7.2%

    c. 3.6% Transformation efficiency (TE) equation: TE = Colonies/µg/Dilution Colonies = the number of colonies counted on the plate µg = the amount of DNA transformed expressed in µg Dilution = the total dilution of the DNA before plating given Colonies = 90 µg DNA = 0.5 mg or 500 micrograms Dilution = 50/1000=0.05 TE = 90/500/0.05 = 3.6 cfu/µg
