Gram staining is a common technique used to differentiate two large groups of bacteria based on their different cell wall constituents. Gram positive bacteria stain violet due to the presence of a thick layer of peptidoglycan in their cell walls and stain react with peptidoglycan forming strong bond which not be easily dissociated. when we wash mixture of gram positive and gram negative bacteria only gram negative bacteria leaves the colour because less stable bond. note important thing that saffranin also react with cell wall component bit when cells already stained with crystal violet stain can not absorb saffranin. Because gram positive bacteria cell wall components are tightly bind with crystal violet.
