A precise definition of flexibility in a particular system requires determination of the number and geometry of the various conformers, their stability, the energy barriers separating the conformers, the kinetic parameters of interconversion and the thermal motion of the atoms within each conformer. Peptide conformational dynamics are important during catalysis, and a balance between rigidity and flexibility means that the peptide can adapt to stabilize the structures of transition states and intermediates throughout the catalytic cycle while simultaneously providing a defined chiral environment, thus enhancing The most common one is to change the amino acid sequence. Branched and bulky amino acids render a peptide more rigid while smaller amino acids make it more flexible. Glycine is a very unique amino acid in that in contains a hydrogen as its side chain (rather than a carbon as is the case in all other amino acids). This means that there is much more conformational flexibility in glycine.
