Split ratio is used during seeding of new cell culture flasks and is based upon growth characteristics of the specific cell line in animal tissue culture. it is determined by confluency. Confluency is important to obtain maximum yeild. In short by using this terminology we can obtain healthy cell culture which bears large size cell than that of mother culture. it defines that either the cell culture is monolatered or not. In simple language we can say that once the cells become 100% confluent the adherent surface of the culture is completely covered by the large healthy cells and any more growth may result in stacking of the cells which will ultimately results in decrease cell culture growth.

how can we determine the split ratio by knowing the confluency of sample ?

as the confluency of culture reaches to 100% further dividing cells gets overlap which re3in decrease in cell surface area and the uptake of minerals by cell decreases.
