    Endonucleases cleave the nucleotide sequence from the middle. Endonuclease cleave the phosphodiester bond present within a polynucleotide chain. Exonucleases cleave DNA sequences in a polynucleotide chain from either the 5' or 3' end one at a time. Means Endonucleases cleave the phosphodiester bond in the polynucleotide present internal in the polynucleotide chain, whereas exonucleases cleave the phosphodiester bond from the ends.
